Design Notes

Ⅰ The last base at the 3' end of the primer is selected as C or G. Ⅱ Tm value of the primer should be controlled between 55℃ and 65℃. Ⅲ The GC content of primers should be controlled between 40% and 60%. Ⅳ Forward and reverse primer Tm values and GC content should be as consistent as possible. Ⅴ The length of primer should be no more than 42bp generally. Ⅵ △Tm should be limited to 2℃
